Studies on the Mechanism of Heat Production by Tissue-Extract

Abstract
The experiment was carried out to ascertain the mechanism of heat production accompanied with the process of tissue necrosis.
1) Normal rabbits were injected intravenously with saline extracts of:
a.)The experimentally necrotized tissues of the liver, spleen and kidneys of the rabbits, ligated at their arteries.
b) The experimentally autolyzed tissues of the heart, liver, spleen and kidney, excised from the rabbits after sacrifice by exsangulpation.
c) The normal tissues of the liver, spleen and kidney, excised front the rabbits after sacrifice by exsanguination.
Examining their fever responses the author found out that in the pyrogenic activity, the necrotize tissue
extract was most active, next came the autolized t. Sue extract and the normal tissuz; extract was active moderately.
2) With successive daily injections of the saline extracts of the autolyzed tissues of the normal rabbits,
the author observed whether tolerance could be obtained by the inoculated animals. But it was concluded that tolerance could not be obtained by them.
3) In order to ascertain Whether the tissue extracts act directly on the thermoreflulatory centers or on the and 6=3 a act i_-directly on them with secondary p5rogendc substance released into the blood stream, normal WIN were injected with sera from the blood of donor rabbits given saline tissue extracts intravenous¢¥ y, and the au or observed their fever responses,
The results showed that they did not release enogenous pyrogenic into the blood stream.
4) The saline extracts of the autolyzed tissues into the
a) The animals (function of the thermoregulatory cecnter inhibited by barbital sodium)
b) The animals (lower cervical section)
c) The animals (bilaterally splanchloctomized t the level just below the diaphragm
d The animals {bilaterally vagotomized at the neck)

The experiments proved that normality¡¯s of the thermoregulatory centers and the thyroid gland were not
cessary for the mechanism of heat production of the autolyzed tissue extacts.
It was believed that the mechanism of heat production by the autolyzed tissue extracts was peripheral.
5) In, order to characterize the nature of the pyrogenic substance which was supposed to exist in the saline extracts of the autolyzed tissues, the author analyzed such extracts biochemically by several methods.
The results showed that the pyrogenic substance belonged mainly to globulin fractionof the extracts.
6) Through these experiments, the author believes that the fever accompanied with the process of tissue
necrosis is caused largely by the pyrogenic substance released from the recretizing tissue and that the pyrogenic substance acts on the peripheral thermogulatoy mechanism or on the fever producing system directly, not through the central thermoregulatory mechanism.
